为研究豆粕替代鱼粉对乌鳢生长、肠道菌群组成和微生物相关氨基酸代谢酶活性的影响,分别用豆粕替代0、25%、50%和75%的鱼粉配制4组等氮等能的饲料饲喂初始体质量为(8.65±0.25)g的幼鱼(分别命名为G1、G2、G3和G4组),养殖实验在室内纤维玻璃钢桶中进行,实验周期为21 d。结果显示,各组间幼鱼成活率无显著性差异;G4组乌鳢幼鱼的生长性能显著低于G1和G2组,而G3组幼鱼生长性能与其余3组差异不显著。厚壁菌门在G1和G2组乌鳢肠道中的比例显著高于G3组,且在G4组中的比例最低;变形菌门在G1和G2组中的比例显著低于G3和G4组;G4组中乳球菌属、杆菌属、假单胞菌属、链球菌属、芽孢杆菌属和不动杆菌属的比例显著低于其余3组。另外,G4组幼鱼肠道微生物谷丙转氨酶、腺苷脱氨酶活性显著高于其余3组,而G1组乳酸脱氢酶的活性最高。研究表明,豆粕替代鱼粉除了对乌鳢幼鱼的生长产生影响外,也显著影响了肠道微生物组成及微生物相关氨基酸代谢酶的活性,本研究结果对于从微生态角度进一步理解豆粕在乌鳢配合饲料中的应用提供了理论依据和参考。  相似文献

Differences in metabolism (enzyme activities, metabolites) between diploid and triploid Salmo trutta f. lacustris were investigated under acclimation and stress conditions. Under acclimation conditions enzyme activities differed for 35% of the 27 investigated key regulatory enzymes and temperature optima for 23%. Muscle and liver metabolites related to energy metabolism and diagnostic indices of blood serum were similar, with exception of acetyl‐CoA being increased in triploids. Metabolic rate was lower and gill ventilation rate higher in triploids in comparison with diploids. During the tested stress situations (24 hr endurance swimming, 3 hr exposure to hypoxia in water with 32% oxygen saturation) muscle and liver glycogen decreased and serum and muscle lactate increased in both ploidy levels. Specifically, for triploids muscle adenylate energy charge and phosphocreatine levels decreased after endurance swimming and muscle and liver adenylate energy charge after exposure to hypoxia. Acetyl‐CoA increased in triploids during both stress situations. In summary, there existed differences in metabolism between the two ploidy levels and the energy metabolism of triploids was less balanced under stress.  相似文献

Marine actinobacteria are viewed as a promising source of enzymes with potential technological applications. They contribute to the turnover of complex biopolymers, such as pectin, lignocellulose, chitin, and keratin, being able to secrete a wide variety of extracellular enzymes. Among these, keratinases are a valuable alternative for recycling keratin-rich waste, which is generated in large quantities by the poultry industry. In this work, we explored the biocatalytic potential of 75 marine-derived actinobacterial strains, focusing mainly on the search for keratinases. A major part of the strains secreted industrially important enzymes, such as proteases, lipases, cellulases, amylases, and keratinases. Among these, we identified two streptomycete strains that presented great potential for recycling keratin wastes—Streptomyces sp. CHA1 and Streptomyces sp. G11C. Substrate concentration, incubation temperature, and, to a lesser extent, inoculum size were found to be important parameters that influenced the production of keratinolytic enzymes in both strains. In addition, proteomic analysis of culture broths from Streptomyces sp. G11C on turkey feathers showed a high abundance and diversity of peptidases, belonging mainly to the serine and metallo-superfamilies. Two proteases from families S08 and M06 were highly expressed. These results contributed to elucidate the mechanism of keratin degradation mediated by streptomycetes.  相似文献

The present study was conducted to investigate the effects of Bacillus pumilus on the growth performance, innate immunity and digestive enzymes of the giant freshwater prawn, Macrobrachium rosenbergii. Prawns (mean weight: 1.81 ± 0.01 g) were fed different levels of Bpumilus at doses of 0, 1 × 107, 1 × 108 and 1 × 109 CFU/g to produce one control group and three experimental groups (G1, G2 and G3), respectively. After a 60‐day experimental feeding period, the results showed that the final weight, weight gain rate (WGR) and specific growth rate (SGR) were significantly higher in experimental groups than in the control (p < 0.05), whereas these improvements did not differ significantly (p > 0.05) among experimental groups. The improvements in phagocytic activity, respiratory burst (RBs) activity and the activity of catalase (CAT), nitric oxide synthase (NOS) and acid phosphatase (ACP) were significantly higher in G2 and G3 than in the control and G1 (p < 0.05). Further, the activity of CAT, NOS and ACP significantly increased with an increase in the dietary level of B. pumilus (p < 0.05). The activity of phenoloxidase (PO) in G3 was significantly higher than that in the control (p < 0.05). The activity of amylase was significantly higher in the groups G2 and G3 than that in the control group. G3 improved the activity of protease compared with the control (p < 0.05). In conclusion, the results of this study indicated that the potential use of B. pumilusat a dose of 1 × 108 CFU/g feed could improve the growth, immunity and digestive enzymes of M. rosenbergii.  相似文献

分析氮对胶东卫矛氮代谢生理的影响规律,明确氮肥施用后的相关酶活性变化机理,为其育苗中科学施用氮肥提供理论依据。试验在田间条件下,设置氮肥施用量分别为0 g/株(J1,CK)、9 g/株(J2)、18 g/株(J3)、27 g/株(J4)4个处理,3次重复。结果表明:5—9月J3谷草转氨酶活性分别比J1提高了64.84%、70.77%、58.52%、47.33%、57.20%,J4与J3之间无显著差异;6—9月J3谷丙转氨酶活性分别比J2提高了18.00%、30.79%、16.47%、16.83%,差异显著;谷胺酰胺合成酶活性7、9月J3显著高于J2,5—6月J2、J3、J4之间无显著差异,J2显著高于对照;5—9月J3硝酸还原酶活性分别比J2提高了20.47%、31.66%、27.32%、23.50%、34.90%,差异显著,J3与J4之间无显著差异;5—7月J3硝态氮含量分别比对照提高了73.76%、53.90%、85.95%,差异显著,8—9月J2、J3、J4之间无显著差异;6—8月,J3可溶性蛋白含量分别比J2提高了15.90%、20.09%、9.04%,差异显著。综合分析认为,J3处理可以显著提高胶东卫矛氮代谢相关酶活性,提高植株叶片内硝态氮含量和可溶性蛋白含量,效果优于J2、J4处理。  相似文献

为探讨龙葵(Solanum nigrum L.)耐镉(Cd)机理,采用基质培养法,研究了50 μmol·L-1 Cd(CdCl2·2.5H2O)处理120 d对其生长和叶片细胞壁果胶、木质素含量、多聚半乳糖醛酸酶(PG)、纤维素酶(Cx)、NADH-过氧化物酶(NADH-POD)、愈创木酚过氧化物酶(GPOD)和松柏醇过氧化物酶(CAPX)活性以及质外体汁液中过氧化氢(H2O2)含量和GPOD、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)和超氧化物歧化酶(SOD)活性的影响。结果表明,与对照相比,Cd处理显著抑制了龙葵地上和地下部的伸长生长、降低了根的干物质积累(P<0.05),但茎的分枝增多,叶面积、叶绿素含量、净光合速率和胞间CO2浓度无显著变化(P>0.05);细胞壁结合酶NADH-POD、GPOD和CAPX活性以及果胶含量显著增高(P<0.05),而PG活性显著下降(P<0.05),Cx活性和木质素含量无显著变化(P>0.05);质外体汁液中H2O2含量显著增加(P<0.05),GPOD和SOD同工酶谱及活性无显著变化(P>0.05);CAT同工酶谱带增宽,APX同工酶谱带数增加,二者活性均显著升高(P<0.05)。说明50 μmol·L-1 Cd较长时间处理龙葵植株,其地上部分生长仍处于良好状态,表现出对Cd毒害的耐性,这与Cd处理后叶片细胞壁果胶含量和结合酶GPOD以及质外体汁液中APX、CAT活性升高有关,推测这是龙葵耐Cd的一种机理。  相似文献

The effects of three treatments of fibrolytic enzymes (cellulase from Trichoderma longibrachiatum (CEL), xylanase from rumen micro‐organisms (XYL) and a 1:1 mixture of CEL and XYL (MIX) on the in vitro fermentation of two samples of Pennisetum clandestinum (P1 and P2), two samples of Dichanthium aristatum (D1 and D2) and one sample of each Acacia decurrens and Acacia mangium (A1 and A2) were investigated. The first experiment compared the effects of two methods of applying the enzymes to forages, either at the time of incubation or 24 h before, on the in vitro gas production. In general, the 24 h pre‐treatment resulted in higher values of gas production rate, and this application method was chosen for a second study investigating the effects of enzymes on chemical composition and in vitro fermentation of forages. The pre‐treatment with CEL for 24 h reduced (p < 0.05) the content of neutral detergent fibre (NDF) of P1, P2, D1 and D2, and that of MIX reduced the NDF content of P1 and D1, but XYL had no effect on any forage. The CEL treatment increased (p < 0.05) total volatile fatty acid (VFA) production for all forages (ranging from 8.6% to 22.7%), but in general, no effects of MIX and XYL were observed. For both P. clandestinum samples, CEL treatment reduced (p < 0.05) the molar proportion of acetate and increased (p < 0.05) that of butyrate, but only subtle changes in VFA profile were observed for the rest of forages. Under the conditions of the present experiment, the treatment of tropical forages with CEL stimulated their in vitro ruminal fermentation, but XYL did not produce any positive effect. These results showed clearly that effectiveness of enzymes varied with the incubated forage and further study is warranted to investigate specific, optimal enzyme‐substrate combinations.  相似文献

为探明耕作方式对豫西旱地麦-豆轮作田不同土层土壤理化特性和土壤酶活性的影响,设置免耕(NT)、旋耕(RT)和翻耕(PT)3种耕作方式,分析了定位试验9 a(2009—2019年)后小麦收获期的土壤容重、孔隙度和有机质、全氮、有效磷和速效钾含量,以及土壤脲酶、蛋白酶、转化酶和碱性磷酸酶活性,并运用相关分析和通径分析研究了土壤理化性质与土壤酶活性的关系。结果表明:不同耕作方式对土壤理化特性和酶活性具有显著的调控作用,其调控效应与其作业深度相关。RT较PT有利于降低0~15 cm土层但增加15~35 cm土层容重,NT较RT和PT显著降低了0~5、15~35、35~60 cm土层的土壤容重,土壤孔隙度变化规律与容重相反。各处理间土壤有机质、全氮、有效磷含量以及土壤脲酶、蛋白酶、转化酶和碱性磷酸酶活性在0~5 cm土层和15~35 cm土层规律一致,0~5 cm土层均以NT最优,分别较最低值处理(PT)显著提高16.7%、53.2%、15.9%、23.6%、18.0%、34.7%和29.0%;15~35 cm土层均以PT最优,分别较最低值处理(NT)显著提高18.3%、23.4%、53.4%、...  相似文献
